Victor John Mature nacque il 29 gennaio 1913 a Louisville, Kentucky, da padre italiano originario di Pinzolo e madre statunitense di origine svizzera. Studiò alla St. Xavier High School, al Kentucky Military Institute e alla Spencerian Business School. Dopo aver svolto vari lavori, si trasferì in California e studiò recitazione al Pasadena Playhouse. Fu scoperto da Charles R. Rogers e firmò un contratto con Hal Roach nel 1939. Debuttò nel film The Housekeeper’s Daughter e ottenne notorietà con One Million B.C. (1940). Fu prestato a RKO per No, No, Nanette e recitò in Captain Caution. Nel 1941 interpretò Randy Curtis nel musical Lady in the Dark a Broadway. Successivamente firmò con 20th Century Fox e recitò in I Wake Up Screaming, The Shanghai Gesture e Song of the Islands. Durante la Seconda Guerra Mondiale si arruolò nella Guardia Costiera degli Stati Uniti e servì a bordo della USCGC Storis. Dopo la guerra, tornò al cinema con My Darling Clementine (1946), Kiss of Death (1947), Fury at Furnace Creek (1948) e Cry of the City (1948).

Nel 1949 interpretò Samson in Samson and Delilah di Cecil B. DeMille, film che incassò oltre 12 milioni di dollari. Recitò in Million Dollar Mermaid (1952), The Robe (1953), Demetrius and the Gladiators (1954), The Egyptian (1954) e Chief Crazy Horse (1955). Firmò contratti con Columbia Pictures, United Artists e Warwick Productions, partecipando a Safari, Zarak, The Sharkfighters, Interpol, The Long Haul, China Doll, Escort West, Timbuktu, The Big Circus e The Bandit of Zhobe. Interpretò Annibale in Hannibal (1959) e Oleg in The Tartars (1962). Si ritirò nel 1966, tornando brevemente per After the Fox (1966), Head (1968), Every Little Crook and Nanny (1972), Won Ton Ton (1976) e Firepower (1979). L’ultima apparizione fu nel film TV Samson and Delilah (1984). Si sposò cinque volte e ebbe una figlia, Victoria, nata nel 1975. Morì il 4 agosto 1999 a Rancho Santa Fe, California, per leucemia, e fu sepolto a Louisville. Ha una stella sulla Hollywood Walk of Fame.
